Analytical Strategy to Monitor Surfactant Integrity by LC-Based Methods and Orthogonal Particle Characterization

Surfactants such as polysorbates and poloxamers are essential excipients in biopharmaceutical formulations, protecting therapeutic proteins from interfacial stress and aggregation. However, hydrolytic and oxidative degradation can compromise their performance, potentially leading to particle formation and reduced product stability.

In this poster, Coriolis Pharma presents an integrated analytical workflow combining liquid chromatography with charged aerosol detection (LC-CAD) and orthogonal particle characterization. The approach enables comprehensive monitoring of surfactant concentration, composition, and degradation pathways while correlating these changes with visible and subvisible particle formation.
Using a case study on enzymatically degraded surfactants, the authors demonstrate how combining LC-based analytics with particle characterization provides a more complete understanding of surfactant integrity and formulation risk. The results highlight the value of a phase-appropriate analytical strategy to support formulation development, comparability assessments, stability studies, and lifecycle management of biotherapeutics.
